目的:观察美满霉素对蒙古沙鼠短暂性脑缺血再灌后前额皮质中钙视网膜蛋白Calretinin(CR)表达的影响,为进一步研究其生理功能及治疗提供参考。方法:42只健康雄性蒙古沙鼠随机分为:正常组对照组、缺血再灌组,美满霉素治疗组。夹闭蒙古沙鼠双侧颈总动脉10 min诱导前脑缺血后,动物分别存活1天、3天或7天。用免疫组化方法检测前额皮质中CR的表达,用图像分析仪测定灰度值。结果:与正常组相比,缺血再灌组与美满霉素治疗组中前额皮质中CR表达均先减少后升高再下降。缺血再灌1天组,CR表达显著减少(P<0.05);3天组CR表达达到高峰(P<0.05);7天组CR表达减弱,但仍高于正常组(P<0.05)。美满霉素治疗各组中的CR表达均弱于对应的缺血再灌组(P<0.05)。结论:美满霉素能抑制蒙古沙鼠脑缺血再灌注后脑皮质中CR的表达来发挥脑保护作用。
Objective: To observe the effect of minocycline on the expression of calretinin (CR) in the prefrontal cortex of Mongolian gerbils after transient forebrain ischemia and provide a reference for further study of its physiological function and treatment. Methods: Forty-two healthy Mongolian gerbils were randomly divided into normal control group, ischemia-reperfusion group and minocycline-treated group. Animals were sacrificed for 1 day, 3 days or 7 days respectively after occlusion of bilateral common carotid arteries of Mongolian gerbils for 10 min. The expression of CR in prefrontal cortex was detected by immunohistochemistry and the gray value was measured by image analyzer. Results: Compared with the normal group, the expression of CR in the prefrontal cortex in ischemic reperfusion group and minocycline group decreased firstly, then increased then decreased. The expression of CR was significantly decreased in the first day after ischemia / reperfusion (P <0.05). The expression of CR was peaked in the third day (P <0.05). The expression of CR in the seventh day was weaker than that in the normal group (P <0.05). Minocycline treatment in each group of CR expression were weaker than the corresponding ischemic reperfusion group (P <0.05). Conclusion: Minocycline can inhibit the expression of CR in cerebral cortex of Mongolian gerbil after cerebral ischemia-reperfusion to exert its neuroprotective effect.
