目的:研究丹参酮ⅡA对人喉癌细胞Hep-2的生长抑制作用及凋亡诱导作用。方法:四亚基噻唑蓝(MTT)法测定丹参酮ⅡA对细胞生长的抑制作用,流式细胞仪技术(FCM)观察细胞周期及凋亡。结果:不同浓度的丹参酮ⅡA均能抑制人喉癌细胞Hep-2生长,并有明显的时间和剂量依赖性;流式细胞仪检测不同浓度丹参酮ⅡA作用48h后的细胞凋亡率增高,与对照组比较差异均有统计学意义。结论:丹参酮ⅡA在体外能明显抑制人喉癌细胞Hep-2生长,抑制其生长的机制可能是诱导细胞凋亡。
Objective: To study the inhibitory effect of tanshinone ⅡA on human laryngeal carcinoma cell line Hep-2 and the induction of apoptosis. Methods: Tetracycline thiazolyl blue (MTT) assay was used to determine the inhibitory effect of tanshinone Ⅱ A on cell growth. Cell cycle and apoptosis were observed by flow cytometry (FCM). Results: Tanshinone ⅡA could inhibit the growth of human laryngeal carcinoma Hep-2 cells in a dose-and time-dependent manner. The apoptosis rate of Tanshinone ⅡA treated with different concentrations of tanshinone ⅡA for 48h was higher than that of the control Group differences were statistically significant. Conclusion: Tanshinone Ⅱ A can inhibit the growth of human laryngeal carcinoma cell Hep-2 in vitro and the mechanism of its inhibition may be to induce cell apoptosis.
