目的初步估计大蒜素与B7.1转基因肿瘤疫苗对膀胱肿瘤的联合抗肿瘤效果。方法采用脂质体将B7.1基因导入鼠膀胱肿瘤(MBT-2)细胞。B7.1分子的表达经免疫荧光染色而测得。利用混合淋巴细胞培养MTT法,观察MBT-2-B7.1细胞刺激脾淋巴细胞的增殖作用。用MTT法测定了大蒜素对肿瘤细胞体外生长的影响。通过动物实验测定该肿瘤疫苗与大蒜素对膀胱肿瘤的联合治疗效果。结果转染了B7.1基因的肿瘤细胞能有效地刺激脾淋巴细胞增殖。当与大蒜素联合应用时,明显延迟了肿瘤的发生,并有效抑制了肿瘤的生长。结论大蒜素与转基因肿瘤疫苗治疗相结合能显著增强抗肿瘤效果。
Objective To estimate the combined anti-tumor effect of allicin and B7.1 transgenic tumor vaccine on bladder cancer. Methods B7.1 gene was transfected into murine bladder tumor (MBT-2) cells by liposome. The expression of B7.1 molecule was measured by immunofluorescence staining. Using mixed lymphocyte culture MTT method to observe the proliferation of splenic lymphocytes stimulated by MBT-2-B7.1 cells. The effect of allicin on the growth of tumor cells in vitro was determined by MTT assay. Through animal experiments to determine the tumor vaccine and allicin on bladder cancer combined treatment. Results B7.1 gene transfected tumor cells can effectively stimulate the proliferation of splenic lymphocytes. When used in combination with allicin, the tumor was significantly delayed, and effectively inhibited the growth of the tumor. Conclusion The combination of allicin and transgenic tumor vaccine can significantly enhance the anti-tumor effect.
