7只正常性周期的奶牛在动情期第17天施行剖腹术,暴露生殖器官。与黄体同侧的子宫动脉周围装电磁流量传感器(5只)。用聚乙烯导管分别经卵巢静脉子宫枝到卵巢静脉(7只)、卵巢动脉子宫枝(5只)和面动脉(7只)行插管术。在第18天,每隔30分钟从三个插管同时采血样。自给雌二醇-17β(E_2)前1小时至给后12小时连续监测子宫血流(E_2给法:ETOH:0.9%NaCl〔50∶50〕溶液6 ml中含3mgE_2,自颈静脉注入)。测定未提取的血浆样品(0.05,0.1或0.2ml)中PGF_(2α)和15-酮-13,14双氢-PGF_(2α)(PGFM)浓度。按Eley等叙述的方法进行PGFM的聚(乙)二醇类放免测定。采用PGF_(2α)抗体测定法。这种抗体在黄体部位与牛血清白蛋白结合。数据按统计学分析系统的普通线性模型程序。通过最小二乘方方差分析
Seven normal-cycle cows were exposed to genital organs on the 17th day of estrus. Peripheral uterine artery with corpus luteum with magnetic flow sensor (5). Polyethylene catheters were used for intubation through ovarian uterine branches to ovary veins (7), ovarian uterine branches (5) and facial arteries (7) respectively. On day 18, blood samples were simultaneously taken from three cannulas every 30 minutes. Uterine blood flow was monitored one hour prior to administration of estradiol-17β (E_2) and 12 hours post-administration (3 mgE_2 in 6 ml of E_2 to ethanol: 50% solution of ETOH: 0.9% NaCl). The concentrations of PGF_ (2α) and 15-keto-13,14 dihydrogen-PGF_ (2α) (PGFM) in non-extracted plasma samples (0.05, 0.1 or 0.2 ml) were determined. The poly (ethylene glycol) radiolysis assay of PGFM was performed as described by Eley et al. PGF 2α antibody assay was used. This antibody binds to bovine serum albumin in the corpus luteum. Data were statistically analyzed for the system’s general linear model program. By least square analysis of variance
