目的:研究运动前补充罗汉果甜苷(Mog)对力竭运动大鼠胃组织热休克蛋白70(HSP70)mRNA表达和HSP70蛋白表达的影响,以初步探讨罗汉果甜苷的作用机制。方法:SD雄性大鼠50只,随机分为5组,每组10只,分别为安静组(NC),运动组(ME),运动+低剂量罗汉果甜苷组(Mog L),运动+中剂量罗汉果甜苷组(Mog M),运动+高剂量罗汉果甜苷组(Mog H)。每天在运动前30 min ig 1次,连续ig 6周,6 d/周,低、中、高3组的ig剂量分别为100,200,400 mg·kg-1,对照组ig等量生理盐水。6周力竭训练结束后,宰杀大鼠。根据试剂盒的方法测定大鼠胃组织HSP70mRNA及蛋白表达。结果:运动组大鼠胃组织HSP70mRNA表达和HSP70蛋白表达水平高于安静组(P<0.05);Mog L,Mog M,Mog H各组大鼠胃组织HSP70mRNA表达和HSP70蛋白水平都高于安静组和运动组(P<0.05,P<0.01)。结论:罗汉果甜苷能够提高大鼠胃组织HSP70mRNA表达和HSP70蛋白表达水平,对减少胃组织的溃疡、促进损伤后的恢复、细胞生长和发育等方面发挥重要的作用。
Objective: To study the effect of mog before exercise on heat shock protein 70 (HSP70) mRNA expression and HSP70 protein expression in gastric tissue of rats after exhaustive exercise to explore the mechanism of mogroside. Methods: Fifty male Sprague-Dawley rats were randomly divided into 5 groups (n = 10): Quiet group (NC), exercise group (ME), Mog L + Mog M and Mog H were used in exercise + high dose mogroside. Ig 1 times daily for 30 min before exercise, ig for 6 weeks, 6 d / week for ig, and 100,200,400 mg · kg-1 for low, middle and high dose groups. The control group was given the same amount of normal saline. After 6 weeks of exhaustive training, the rats were slaughtered. The expression of HSP70mRNA and protein in gastric tissue of rats were determined by kit method. Results: The expression of HSP70 mRNA and HSP70 protein in gastric tissue of exercise group were higher than that of quiet group (P <0.05). The expression of HSP70 mRNA and HSP70 protein in gastric tissue of Mog L, Mog M and Mog H groups were higher than that of quiet group And exercise group (P <0.05, P <0.01). Conclusion: Mangosteen glycosides can improve the expression of HSP70mRNA and HSP70 protein in gastric tissue of rats, and play an important role in reducing gastric ulcer, promoting the recovery after injury, cell growth and development.
