目的 探讨Bax表达变化与PPAR γ激动剂诱导的肺癌细胞凋亡的关系。方法 以RT PCR和Westernblot分别检测肺癌细胞株上PPAR γ的表达 ,并通过TUNEL法检测经PPAR γ的激动剂 (配体 )诱导的细胞凋亡 ,经原位分子杂交技术和免疫组织化学方法检测诱导凋亡前、后Bax的mRNA和蛋白水平表达的变化。结果 肺癌细胞上有PPAR γ的表达 ,且PPAR γ的激动剂能抑制肺癌细胞生长并诱导细胞凋亡 ,Bax在诱导调亡后表达较前增加 ,且与诱导凋亡的程度呈正相关。结论 Bax作为一种促凋亡因子 ,在PPAR γ的激动剂诱导的肺癌细胞调亡中起作用。
Objective To investigate the relationship between the changes of Bax expression and the apoptosis of lung cancer cells induced by PPARγ agonist. Methods The expression of PPARγ in lung cancer cell lines was detected by RT PCR and Western blot respectively. Apoptosis induced by PPARγ agonist (ligand) was detected by TUNEL assay. The expression of PPARγ was detected by in situ hybridization and immunohistochemistry Changes of Bax mRNA and protein expression before and after induction of apoptosis. Results The expression of PPARγ was found in lung cancer cells, and PPARγ agonist could inhibit the growth of lung cancer cells and induce apoptosis. The expression of Bax in the lung cancer cells increased compared with the former, and positively correlated with the degree of apoptosis induction. Conclusion Bax as a pro-apoptotic factor plays a role in PPARγ-induced apoptosis of lung cancer cells.
