研究干扰素α(IFN-α)和肿瘤坏死因子α(TNF-α)对乳腺癌耐药细胞株MCF7/ADR耐药性的逆转作用,并探讨其对多药耐药基因mdrl和谷胱甘肽-S-转移酶(GSTπ)表达以及细胞内ADR积累的影响,我们应用细胞培养和MTT法,分析应用IFN-α和TNF-α(均为100 U/ml)处理前后,MCF7/ADR对ADR敏感性的变化,以ADR的半数抑制浓度(IC_(50))作为评价敏感性的指标.采用半定量RT-PCR和免疫组化染色法,观察药物处理后不同时间mdrl和GSTπ的mRNA和蛋白表达的变化规律;应用流式细胞技术检测MCF7/ADR细胞内ADR浓度的变化.结果发现,MCF7/ADR对ADR的IC_(50)为30
To study the reversal effect of interferon-alpha (IFN-alpha) and tumor necrosis factor-alpha (TNF-alpha) on the drug resistance of breast cancer resistant cell line MCF7/ADR and to investigate its multidrug resistance gene mdrl and glutathione The effect of peptide-S-transferase (GSTπ) expression and intracellular ADR accumulation, we used cell culture and MTT assay to analyze the application of IFN-α and TNF-α (both 100 U/ml) before and after MCF7/ADR treatment. Changes in protein expression; Flow cytometry was used to detect changes in ADR concentrations in MCF7/ADR cells. The results showed that MCF7/ADR had an IC 50 of ADR of 30.
