目的研究TSC2基因的表达产物tuberin在结直肠癌中mRNA和蛋白水平的表达及其与临床病理特征的关系,探讨其在结直肠癌发生中的作用。方法采用实时荧光定量逆转录聚合酶链反应方法检测40例结直肠癌与癌旁正常肠黏膜组织中tuberin mRNA的表达,应用免疫组织化学方法检测60例结直肠癌与癌旁正常肠黏膜组织tuberin蛋白的表达,分析tuberin mRNA和蛋白表达与临床病理特征的关系。结果40例结直肠癌中tuberin mRNA表达量与患者年龄、性别、肿瘤大小、组织学分级、临床分期、淋巴结转移、远处转移无相关性(P>0.05),但显著低于癌旁正常组织,差异有高度统计学意义(P﹤0.01)。tuberin蛋白在60例结直肠癌与癌旁正常肠黏膜组织中的阳性表达率分别为41.6%、75%,差异有统计学意义(P﹤0.05)。tuberin蛋白阳性表达率与性别、组织学分级相关。结论tuberin在结直肠癌中表达下调,可能参与结直肠癌的发生。
Objective To investigate the expression of tuberculin (TSC2) mRNA and protein in colorectal cancer and its relationship with clinicopathological features and to explore the role of TSC2 gene in colorectal cancer. Methods The expression of tuberin mRNA in 40 colorectal cancer tissues and adjacent normal mucosa tissues was detected by real-time fluorescence quantitative reverse transcription polymerase chain reaction (RT-PCR). Immunohistochemistry was used to detect the expression of tuberin in 60 colorectal cancer tissues and adjacent normal mucosa tissues. Protein expression, analyze tuberin mRNA and protein expression and clinicopathological features. Results The expression of tuberin mRNA in 40 cases of colorectal cancer had no correlation with the age, gender, tumor size, histological grade, clinical stage, lymph node metastasis and distant metastasis (P> 0.05), but significantly lower than that in the adjacent normal tissues , The difference was highly statistically significant (P <0.01). The positive rates of tuberin protein in 60 cases of colorectal cancer and adjacent normal mucosa tissues were 41.6% and 75%, respectively, with significant difference (P <0.05). The positive rate of tuberin protein was related to sex and histological grade. Conclusion The expression of tuberin is down-regulated in colorectal cancer and may be involved in colorectal cancer.
