目的 探讨IL 4、紫杉醇对Lewis肺癌细胞增殖的影响。方法 将Lewis肺癌细胞 ( 1 2 5×10 6个 /鼠 )接种于C57BL/6小鼠皮下。将 40只小鼠随机分为Ⅰ、Ⅱ、Ⅲ、Ⅳ组 ,接种后第 5天起分别给予生理盐水、IL 4、紫杉醇和IL 4+紫杉醇。第 18天处死动物。利用图像分析仪对肿瘤细胞核形态、DNA含量及AgNORs计数进行测量。结果 Ⅱ组核面积为 ( 2 4 2 0 2± 5 0 12 ) μm2 ,周长 ( 18 62 5± 4 180 ) μm ,平均直径 ( 2 64 2± 0 2 91) μm ,积分光密度 3 412± 0 910 ,AgNORs计数 5 3 13± 0 82 0 ,各项参数数值均显著小于Ⅰ组 (P <0 .0 5 ) ;Ⅲ组核面积为 ( 18 3 73± 4.83 3 ) μm2 ,周长 ( 16 5 2 2± 2 5 18) μm ,平均直径 ( 2 40 1±0 2 88) μm ,积分光密度 3 0 96± 0 771,AgNORs计数 2 670± 0 40 1,均显著小于Ⅰ组 (P <0 .0 1) ;Ⅳ组核面积为 ( 16 664± 4 671) μm2 ,周长 ( 15 90 1± 2 70 0 ) μm ,平均直径 ( 2 2 83± 0 3 0 8) μm ,积分光密度 2 70 7±0 80 7,AgNORs计数 1 5 5 2± 0 3 16,均显著小于Ⅰ组 (P <0 .0 1)。各用药组间各参数差异亦具有显著性(P <0 .0 5 )。结论 IL 4和紫杉醇对Lewis肺癌细胞的增殖均有抑制作用 ,紫杉醇疗效较IL 4好 ,而两者联用疗效更好。
Objective To investigate the effects of IL 4 and paclitaxel on the proliferation of Lewis lung carcinoma cells. Methods Lewis lung carcinoma cells (125 × 10 6 cells / mouse) were inoculated subcutaneously in C57BL / 6 mice. Forty mice were randomly divided into groups Ⅰ, Ⅱ, Ⅲ and Ⅳ. Normal saline, IL 4, paclitaxel and IL 4 + paclitaxel were given on the 5th day after inoculation. The animals were sacrificed on the 18th day. Tumor cell nuclear morphology, DNA content and AgNORs counts were measured using an image analyzer. Results The nucleus area of group Ⅱ was (2220 ± 5 0 12) μm2, the circumference was (18 62 5 ± 4 180) μm, the average diameter was 2 642 ± 0 2 91 μm, the integral optical density was 31 412 ± 0 910 and AgNORs 533 ± 0 82 0, the values of all parameters were significantly lower than those in group Ⅰ (P <0.05); the area of group Ⅲ was (18 3 73 ± 4.83 3) μm 2 and the circumference 5 22 ± 2 5 18) μm, mean diameter (2 40 1 ± 0 2 88) μm, integral optical density 3 0 96 ± 0 771 and AgNORs 2 670 ± 0 40 1 were significantly lower than those in group Ⅰ (P < 0 1 01). The nucleus area of group Ⅳ was (16 664 ± 4 671) μm 2, the circumference was (15 90 1 ± 2 70 0) μm and the average diameter was (2283 ± 0 308) μm. The integral optical density 2 70 7 ± 0 80 7 and AgNORs 1 5 5 2 ± 0 3 16 were significantly lower than those in group Ⅰ (P <0.01). There was also significant difference in each parameter between the two groups (P <0.05). Conclusions Both IL 4 and paclitaxel can inhibit the proliferation of Lewis lung cancer cells. Paclitaxel is more effective than IL 4 and the combination of both is better.
