目的:运用脉冲场凝胶电泳(PFGE)技术对一起由副溶血性弧菌引起的食物中毒所分离的菌株进行同源性分析。方法:对分离的副溶血性弧菌进行常规鉴定、血清分型后,进行PFGE分析并确定分子分型,用B ioNum erics软件对PFGE分型图谱进行电子化数据分析,绘出聚类分析图,观察引起食物中毒的菌型是否为同一分子分型的菌株引起。结果:此次食物中毒分离到11株副溶血性弧菌,其中从病人粪便中分离10株,刀具涂抹物中分离1株,血清型均为O3:K6型,这些菌株经PFGE聚类分析得到3种带型,3型之间分别有1~2条电泳条带的差异,相似性百分比为93%。结论:此次食物中毒是由同一克隆副溶血性弧菌引起,凝胶电泳技术适用于菌株的同源性的分析和传染源的追踪。
OBJECTIVE: To analyze the homology of strains isolated from food poisoning caused by Vibrio parahaemolyticus by pulsed-field gel electrophoresis (PFGE). Methods: The isolated Vibrio parahaemolyticus was routinely identified. After serotyping, the PFGE analysis was performed to determine the molecular typing. The electronic data analysis of the PFGE typing patterns was performed by Bioinformatics software, and the cluster analysis was drawn , To observe whether the type of food poisoning caused by the same molecular typing strains. Results: 11 strains of Vibrio parahaemolyticus were isolated from the food poisoning. Among them, 10 strains were isolated from the feces of the patients and 1 strain was isolated from the tool spreads. The serotypes of the strains were all O3: K6 strains. These strains were obtained by PFGE cluster analysis There were 1 ~ 2 electrophoretic bands differences between the 3 types of bands and 3 types respectively, with a similarity of 93%. Conclusion: This food poisoning is caused by the same clone of Vibrio parahaemolyticus. The gel electrophoresis technique is suitable for the homologous analysis of strains and the tracking of infectious agents.
