以大岩桐(SinnigiaspeciosaBenth.etHook)叶片为试材,接种于MS附加不同激素组合的培养基上,成功地经愈伤组织亚球状体至不定芽途径形成大量再生植株并移栽成活。在各种细胞分裂素中,6-苄基嘌呤(BA)的效果优于激动素(KT)。生长素萘乙酸(NAA)与BA及KT配合使用时愈伤组织诱导、球状体分化及不定芽增殖均有明显的增效作用。其最适宜的培养基组成为:MS+BA2.0mg/L+NAA0.2mg/L在这个培养基上,愈伤组织诱导率为94%,球状体分化率为188%,不定芽在口周内增殖5倍。最适宜的生极培养基为:MS+NAA0.5mg/L,生根率在90%以上,试管苗移栽成活率在80%以上。
The leaves of Sinnigiaspeciosa Benth.etHook were inoculated on MS medium supplemented with different hormones, and a large number of regenerated plants were successfully formed from the sub-spheroids to adventitious buds of callus and transplanted.
