目的探讨CD47在急性白血的表达及临床意义。方法流式细胞仪检测183例急性白血病患者骨髓CD47的表达,以及聚合酶链反应-变性高效液相色谱检测FLT3-ITD突变,Real-Time PCR检测BCRABL融合基因。分析各亚型急性白血病的CD47表达差异以及CD47阳性与高白细胞,完全缓解率和复发的影响。结果急性白血病CD47较健康对照组明显高表达,AML中,AML-M0,M4和M6表达高,ALL中T细胞较B细胞ALL表达高,CD47的高表达与高白细胞有关,AML中FLT3-ITD突变与CD47无关,但是BALL的BCR-ABL阳性与CD47表达有关,而且CD47阳性白血病患者CR率低,AML患者CD47阳性容易复发。结论 CD47在AL的表达可以判断AL疗效,也是AML复发的一个独立因素。
Objective To investigate the expression of CD47 in acute leukemia and its clinical significance. Methods The expression of CD47 in bone marrow of 183 patients with acute leukemia was detected by flow cytometry. The FLT3-ITD mutation was detected by polymerase chain reaction-denaturing high performance liquid chromatography. The BCRABL fusion gene was detected by Real-Time PCR. To analyze the difference of CD47 expression in each subtype of acute leukemia and the effect of CD47 positive and high white blood cells, complete remission rate and relapse. Results The expression of CD47 in acute leukemia was significantly higher than that in healthy controls. In AML, the expression of AML-M0, M4 and M6 was high, the expression of T cells in ALL was higher than that in B cells, and the expression of CD47 was associated with high leukocytes. FLT3-ITD The mutation is not associated with CD47, but BALL-positive for BALL is associated with CD47 expression, and CR rates are low in patients with CD47-positive leukemia and CD47-positive relapsing in AML patients. Conclusion The expression of CD47 in AL can judge the effect of AL and is also an independent factor of AML recurrence.
