目的:疼痛特别是慢性疼痛,是战争创伤引起的常见症状,也是近年来影响人们身心健康的主要因素之一,但其对消化系统功能的影响仍没有系统的研究。因此,文中通过建立坐骨神经选择性损伤模型研究慢性疼痛应激状态下SD大鼠胃中annexin 5表达的变化。方法:72只SD大鼠随机分成3组,手术组大鼠结扎并切断胫神经和腓总神经,但保留腓肠神经以建立坐骨神经分支选择性损伤(SNI)模型;假手术组只暴露坐骨神经干及其分支而不损伤神经;正常对照组不做任何处理。然后通过免疫组织化学和Western blot方法分别研究坐骨神经选择性损伤后14 d、21 d和28 d时大鼠胃中annexin 5蛋白定位及表达的变化。结果:免疫组化结果显示annexin 5存在于胃上皮及胃腺细胞中。术后14d,手术组胃黏膜上皮细胞中annexin 5的定位明显减少;Western blot结果显示,在坐骨神经分支选择性损伤引起的慢性疼痛应激状态下,14 d时手术组annexin 5蛋白表达水平显著低于对照组(P<0.01)和假手术组(P<0.05),21 d时手术组annexin 5蛋白表达水平也显著低于对照组和假手术组(P<0.05),而48 d时手术组与其对应的对照组和假手术组相比均无显著性差异(P>0.05)。结论:坐骨神经选择性损伤引起的慢性疼痛应激能下调SD大鼠胃中annexin 5的表达,annexin 5可能参与了胃对疼痛应激的反应。
OBJECTIVE: Pain, especially chronic pain, is a common symptom caused by war trauma. It is also one of the major factors affecting people’s physical and mental health in recent years. However, its influence on digestive system function is still not systematically studied. Therefore, we established a selective sciatic nerve injury model to study the changes of annexin 5 expression in the stomach of SD rat under chronic pain stress. Methods: Seventy-two SD rats were randomly divided into three groups. The rats in the operation group were ligated and cut the tibial nerve and common peroneal nerve, while the sural nerve was preserved to establish the selective injury of the sciatic nerve branch (SNI) model. Only the sciatic nerve trunk And its branches without damage to nerves; normal control group without any treatment. Then immunohistochemistry and Western blot were used to investigate the location and expression of annexin 5 protein in rat stomach at 14 d, 21 d and 28 d after sciatic nerve selective injury respectively. Results: The results of immunohistochemistry showed annexin 5 existed in gastric epithelium and gastric gland cells. After 14 days, the location of annexin 5 in gastric mucosal epithelium decreased significantly in the operation group. The Western blot results showed that the annexin 5 protein expression in the operation group was significantly lower at 14 d after chronic pain-induced stress induced by selective sciatic nerve branch injury In the control group (P <0.01) and sham operation group (P <0.05), the expression level of annexin 5 in the operation group was significantly lower than that in the control group and the sham operation group on the 21st day (P <0.05) There was no significant difference between the control group and the sham operation group (P> 0.05). CONCLUSION: Chronic pain stress induced by selective sciatic nerve injury can down-regulate the expression of annexin 5 in the stomach of SD rats, and annexin 5 may be involved in the response of stomach to pain stress.
