目的探讨垂体肿瘤转化基因1(PTTG1)调控碱性成纤维细胞生长因子(bFGF)、基质金属蛋白酶-2(MMP-2)蛋白表达在子宫内膜癌发生、侵袭和转移中的作用及意义。方法采用免疫组化方法,结合组织芯片技术,检测2005年1月至2009年9月上海交通大学附属上海市第一人民医院124例子宫内膜样腺癌、28例内膜不典型增生、35例正常内膜组织中PTTG1、bFGF和MMP-2的表达水平,结合临床病理因素进行分析。结果 PTTG1、bFGF和MMP-2在子宫内膜样腺癌中的阳性表达率分别为63.7%、73.4%、50.8%,与正常内膜组和不典型增生组比较,差异有统计学意义(P<0.01)。PTTG1蛋白与手术病理分期显著相关(P<0.01),bFGF表达与肿瘤肌层浸润和淋巴结转移明显相关(P<0.05),而MMP-2蛋白表达与肿瘤淋巴结转移显著相关(P<0.01)。子宫内膜样腺癌组织中PTTG1和bFGF、MMP-2的表达均存在正相关关系(r=0.262,P=0.003;r=0.360,P<0.001)。结论 PTTG1可能上调bFGF和MMP-2蛋白的表达,在子宫内膜癌侵袭和转移中起重要作用。
Objective To investigate the role and significance of pituitary tumor transforming gene 1 (PTTG1) in regulating the expression of basic fibroblast growth factor (bFGF) and matrix metalloproteinase-2 (MMP-2) in the pathogenesis, invasion and metastasis of endometrial carcinoma. Methods Using immunohistochemistry and tissue microarray technique, 124 cases of endometrial adenocarcinoma, 28 cases of atypical hyperplasia of atypia in Shanghai First People’s Hospital affiliated to Shanghai Jiaotong University from January 2005 to September 2009 were detected. Cases of normal endometrial tissue PTTG1, bFGF and MMP-2 expression levels, combined with clinicopathological factors were analyzed. Results The positive rates of PTTG1, bFGF and MMP-2 in endometrial adenocarcinoma were 63.7%, 73.4% and 50.8%, respectively, which were significantly different from those in normal endometrium and atypical hyperplasia (P <0.01). The expression of PTTG1 protein was significantly correlated with the pathological stage (P <0.01). The expression of bFGF was significantly correlated with myometrial invasion and lymph node metastasis (P <0.05), while the expression of MMP-2 protein was significantly correlated with lymph node metastasis (P <0.01). There was a positive correlation between the expression of PTTG1, bFGF and MMP-2 in endometrioid adenocarcinoma (r = 0.262, P = 0.003; r = 0.360, P <0.001). Conclusions PTTG1 may up-regulate the expression of bFGF and MMP-2 protein and play an important role in the invasion and metastasis of endometrial carcinoma.
