通过Northern斑点杂交技术研究52例颊癌及相关组织中肿瘤转移抑制基因nm23-H1和nm23-H2mRNA的表达。结果发现:颊癌组织中nm23-H1和nm23-H2mRNA表达比正常颊粘膜、白斑、癌旁粘膜均有不同程度增高。颊癌有转移组中nm23-H1mRNA表达比无转移组明显降低,转移灶中nm23-H1mRNA的表达更低(P<0.05)。11例有转移颊癌患者中有9例(81.8%)为nm23-H1低表达;而19例无转移颊癌,有15例(78.9%)为高表达(P<0.05)。nm23-H2mRNA表达在有无转移组患者中无明显差异(P>0.05)。结果提示,在颊癌的转移过程中,nm23-H1比nm23-H2起着更重要的作用。nm23-H1mRNA表达可作为预测有无颊癌淋巴结转移的一个有价值的指标。
The expression of tumor metastasis suppressor genes nm23-H1 and nm23-H2 in 52 cases of buccal carcinoma and related tissues were studied by Northern blotting. The results showed that: the expression of nm23-H1 and nm23-H2mRNA in buccal carcinoma tissues were higher than that in normal buccal mucosa, leukoplakia and adjacent mucosa. The expression of nm23-H1mRNA in buccal carcinoma metastasis group was significantly lower than that in non-metastasis group, and the expression of nm23-H1mRNA in metastasis was lower (P <0.05). Nine of 11 (81.8%) patients with metastatic buccal carcinoma had a low expression of nm23-H1, while 19 had no metastatic buccal carcinoma (15%, 78.9%, P <0.05) ). There was no significant difference in the expression of nm23-H2mRNA between the patients with and without metastasis (P> 0.05). The results suggest that nm23-H1 plays a more important role than nm23-H2 in the metastasis of buccal carcinoma. The expression of nm23-H1mRNA can be used as a valuable index to predict the metastasis of lymphnode with or without buccal carcinoma.
