目的观察艾麻口服液对慢性支气管炎大鼠气道炎症的影响,并探讨其部分疗效机制。方法用气管内注入低剂量脂多糖复制大鼠慢性支气管炎模型。雄性Wistar大鼠随机分为假手术组、慢支模型组、艾麻口服液组及橘红止咳口服液对照组(n=7)。测定各组大鼠支气管肺泡灌洗液(BALF)细胞总数及白细胞分类计数,肺组织中髓过氧化物酶(MPO)活力,BALF、血浆及肺组织匀浆液中总超氧化物歧化酶(SOD)活力和丙二醛(MDA)含量,测定大鼠BALF、肺组织及外周血中肿瘤坏死因子-α(TNF-α)、白细胞介素-8(IL-8)的含量。结果慢性支气管炎大鼠BALF中炎性细胞明显增高,肺组织中的MPO活性、肺组织及外周血中MDA含量明显增加,BALF、肺组织及外周血中总SOD活力则明显降低;经艾麻口服液治疗后,BALF中炎症细胞分类计数、肺组织中MPO活性、肺组织及外周血中MDA含量均明显降低,BALF、肺组织及外周血中总SOD活力明显升高。进一步研究显示,艾麻口服液能减少慢支大鼠BALF中IL-8的含量,减少慢支大鼠BALF、肺组织匀浆液及血浆中TNF-α含量。结论艾麻口服液通过调节TNF-α、IL-8的分泌以减少慢支气道炎性细胞的浸润,同时具有促进氧自由基的清除和抗脂质过氧化损伤作用。
Objective To observe the effect of Aima Oral Liquid on airway inflammation in rats with chronic bronchitis and to explore its partial therapeutic mechanism. Methods Low-dose lipopolysaccharide was injected intratracheally to replicate chronic bronchitis in rats. Male Wistar rats were randomly divided into sham operation group, chronic bronchitis model group, Aima oral liquid group, and Tanghong Hong Zhike oral liquid control group (n=7). The total number of cells and leukocyte counts in the bronchoalveolar lavage fluid (BALF), the myeloperoxidase (MPO) activity in the lung tissue, the total superoxide dismutase (SOD) in the BALF, plasma and lung tissue homogenates were determined. ) Vigor and malondialdehyde (MDA) content were measured in BALF, lung tissue, and peripheral blood tumor necrosis factor-α (TNF-α) and interleukin-8 (IL-8) contents. Results The inflammatory cells in BALF of rats with chronic bronchitis were significantly increased, MPO activity in lung tissue, MDA content in lung tissue and peripheral blood were significantly increased, and the total SOD activity in BALF, lung tissue, and peripheral blood was significantly decreased; After oral liquid treatment, the number of inflammatory cells in BALF, MPO activity in lung tissue, and MDA content in lung tissue and peripheral blood were all significantly reduced, and the total SOD activity in BALF, lung tissue, and peripheral blood was significantly increased. Further studies have shown that Aima oral liquid can reduce the IL-8 content in BALF of chronic bronchitis rats and reduce the content of TNF-α in BALF, lung homogenate and plasma of chronic bronchitis rats. Conclusion Aima oral liquid can reduce the infiltration of inflammatory cells of chronic bronchitis by regulating the secretion of TNF-α and IL-8, and it can also promote the removal of oxygen free radicals and anti-lipid peroxidative damage.
