目的:观察健脾益气摄血方对辛伐他汀诱导斑马鱼出血的止血效果并与清热凉血方比较。方法:用辛伐他汀0.5μmol/L处理AB系斑马鱼24 h,建立斑马鱼出血模型,在严格盲态实验条件下,在最大非致死量范围内,应用不同浓度实验药物处理出血斑马鱼,以出血减少率、止血率为主要观察指标,以类似脾气虚特点的血流量、血流量改善率与运动速度、运动改善率为附加指标,综合评价健脾益气摄血方的干预效果。结果:实验药物B1在浓度为500和1 000μg/ml时出血斑马鱼的出血率分别为30%和15%,止血率分别为60%和80%;实验药物B2在浓度为500和1 000μg/ml时出血斑马鱼出血率分别为45%和40%,止血率分别为40%和47%。表明在降低斑马鱼出血率与止血效果两方面实验药物B1优于实验药物B2。在相等浓度下,在提高出血斑马鱼血流量与血流量改善率两方面实验药物B1优于实验药物B2。在相等浓度下,在促进斑马鱼运动速度与提高斑马鱼运动改善率方面实验药物B1优于实验药物B2。结论:健脾益气摄血方对辛伐他汀诱导的斑马鱼出血有良好的止血效果,并能够改善出血斑马鱼血流量与提高运动速度,为健脾益气摄血方治疗脾不统血证提供了实验依据。
Objective: To observe the effect of invigorating the spleen and replenishing qi on hemostasis induced by simvastatin in hemorrhagic zebrafish and compare with Qingre Liangxue prescription. Methods: The zebrafish was treated with simvastatin 0.5 μmol / L for 24 h to establish a model of zebrafish hemorrhage. Under the condition of maximum non-lethal dose, zebrafish hemorrhage was treated with different concentrations of experimental drugs under strict blind experimental conditions. Hemorrhage reduction rate and hemostasis rate were taken as the main observation indexes. The blood flow, blood flow improvement rate, movement speed and motor improvement rate were similar to those of the spleen deficiency syndrome. Results: The hemorrhage rates of experimental drug B1 in hemorrhagic zebrafish at concentrations of 500 and 1 000 μg / ml were 30% and 15%, respectively, and the rates of hemostasis were 60% and 80% respectively.
