目的 探讨SeO2对三种肺癌肿瘤细胞A549,GLC-82和PG的增生,凋亡,Bcl-2和p53表达,活性氧和Ca2+水平的影响。方法 本实验采用流式细胞术测定了细胞的凋亡率,细胞中活性氧和Ca2+的水平以及Bcl-2和p53的表达。结果 10μmoL/L的SeO2作用48 h就能诱导47.8%的A549和40.8%的GLC-82细胞凋亡,而30 μmol/L的SeO2也能诱导37.8%的PG细胞凋亡。用不同浓度(3-30 μmoL/L)的SeO2处理细胞48 h,在A549细胞中,代表细胞Bcl-2表达的平均荧光强度从65.8下降至9.6,而在其它两种细胞中,Bcl-2的表达下降都不明显。P53的表达在三种细胞中都有不同程度的上升,在A549细胞中,从3.5上升至7.1,在GLC-82细胞中,从4.7上升至7.4,在PG细胞中,从7.8上升至9.0。SeO2能显著降低三种细胞内活性氧和Ca2+水平。结论 SeO2对三种肺癌肿瘤细胞均有诱导凋亡的作用,在凋亡过程中涉及到Bcl-2下调和p53上调,以及细胞内活性氧和Ca2+水平下降。
Objective To investigate the effects of SeO2 on proliferation, apoptosis, expression of Bcl-2 and p53, reactive oxygen species and Ca2 + in three lung cancer cell lines A549, GLC-82 and PG. Methods Flow cytometry was used to determine the apoptosis rate of cells, the levels of reactive oxygen species and Ca2 + in cells, and the expressions of Bcl-2 and p53. Results SeO2 at 10μmol / L induced apoptosis in 47.8% A549 and 40.8% of GLC-82 cells at 48 h, while SeO2 at 30 μmol / L induced apoptosis in 37.8% of PG cells. The average fluorescence intensity of Bcl-2 expression in A549 cells decreased from 65.8 to 9.6 with SeO2 at different concentrations (3-30 μmoL / L) for 48 h, whereas in the other two cells, Bcl-2 The decrease of expression was not obvious. P53 expression increased to varying degrees in all three types of cells, from 3.5 to 7.1 in A549 cells, from 4.7 to 7.4 in GLC-82 cells and from 7.8 to 9.0 in PG cells. SeO2 significantly reduced the levels of reactive oxygen species and Ca2 + in three kinds of cells. Conclusion SeO2 induces apoptosis in all three kinds of lung cancer cell lines, including down-regulation of Bcl-2 and up-regulation of p53, and decrease of intracellular reactive oxygen species and Ca2 + levels during apoptosis.
