目的研究理脾调脂胶囊对血脂失调症大鼠ApoE、ApoCⅢ基因表达的影响,探讨其调节血脂代谢的作用机制。方法 Wistar大鼠随机分为正常组、模型组、理脾调脂胶囊治疗组(简称治疗组)、血脂康对照组(简称对照组),造模4周后,治疗组大鼠每日灌胃理脾调脂胶囊,对照组每日灌胃血脂康,用药8周。实验结束时,酶法测定大鼠血清TC、TG浓度,沉淀法测定大鼠血清HDL-c、LDL-c;实时定量PCR法(Real-time PCR)检测大鼠肝脏ApoE、血浆中ApoCⅢmRNA的表达。结果与正常组比较,模型组大鼠血清中的TC、TG、LDL-c均显著升高(P<0.01),HDL-c降低(P<0.01),ApoEmRNA的表达量降低(P<0.05),血浆ApoCⅢ基因表达显著升高(P<0.01);与模型组比较,治疗组和对照组大鼠血清中TC、TG、LDL-c均降低(P<0.01),治疗组大鼠血清HDL-c升高较为明显(P<0.01),ApoEmRNA的表达量升高较为显著(P<0.01),血浆ApoCⅢ基因表达显著降低(P<0.01);与对照组比较,治疗组大鼠血清中TC、TG、LDL-c均降低(P<0.05),HDL-c升高(P<0.01),血浆ApoCⅢ基因表达显著降低(P<0.01)。结论理脾调脂胶囊能显著提高实验性血脂失调症大鼠肝脏ApoEmRNA的表达,并降低血中ApoCⅢmRNA的表达,具有调控血脂代谢的作用。
Objective To investigate the effect of Lipi Tiaozhi Capsule on the gene expression of ApoE and ApoC Ⅲ in rats with dyslipidemia and to explore its mechanism of action in regulating blood lipid metabolism. Methods Wistar rats were randomly divided into normal group, model group, Li Pi Tiao Zhi Capsule treatment group (referred to as treatment group) and Xuezhikang control group (control group). After 4 weeks of modeling, Lipi Tiaozhi Capsule, the control group were fed Xuezhikang daily, medication for 8 weeks. At the end of the experiment, the levels of serum TC and TG in rat serum were determined by enzyme-linked immunosorbent assay (ELISA), HDL-c and LDL-c were measured by sedimentation method. ApoE mRNA expression in liver was detected by real-time PCR . Results Compared with the normal group, the levels of TC, TG and LDL-c in the model group were significantly increased (P <0.01), HDL-c was decreased (P <0.01), and the expression of ApoE mRNA was decreased (P <0.01). Compared with the model group, the levels of TC, TG and LDL-c in the serum of the treated group and the control group were significantly decreased (P <0.01). The levels of serum HDL- (P <0.01). The expression of ApoE mRNA was significantly increased (P <0.01) and the expression of ApoC Ⅲ gene in plasma was significantly lower (P <0.01). Compared with the control group, the levels of TC, TG and LDL-c decreased (P <0.05), HDL-c increased (P <0.01), plasma ApoCⅢ gene expression decreased significantly (P <0.01). Conclusion Li-spleen Tiaozhi capsule can significantly improve the experimental dyslipidemia rat liver ApoEmRNA expression, and reduce blood ApoC ¢ ómRNA expression, with the regulation of blood lipid metabolism.
