香梨优斑螟(Euzophera pyriella Yang)作为新疆林果的重要害虫,对水果生产和贸易都有一定的影响。其各虫态的形态特征与近缘种相似,不易进行快速鉴定。本研究选择线粒体DNA细胞色素氧化酶I(COI)基因片段作为物种鉴定的条形码,结合BOLD系统的鉴定工具和系统发育树构建,对香梨优斑螟进行了分子鉴定。结果发现检测的9个样本(成虫、蛹、幼虫和卵)全为香梨优斑螟,相似度在99.23%以上,在以NJ方法构建的系统发育树中,明显与近缘种枇杷暗斑螟(Euzopherabigella)区分开。
Euzophera pyriella Yang, an important pest of Xinjiang forest fruit, has certain influence on fruit production and trade. The morphological characteristics of its various insect states and related species similar, not easy to quickly identify. In this study, a mitochondrial DNA cytochrome oxidase I (COI) gene fragment was selected as a barcode for species identification, and molecular identification was performed using the BOLD system identification tool and phylogenetic tree construction. The results showed that 9 samples (adult, pupae, larvae and eggs) were all tested for C. excellense with similarities of more than 99.23%. Among the phylogenetic trees constructed by NJ method, Borer (Euzopherabigella) distinguish.
