目的研究体外培养盆底功能障碍性疾病(pelvic floor dysfunction,PFD)患者阴道壁成纤维细胞的力学反应及17β-雌二醇(E2)对其力学反应的影响。方法河北医科大学第二医院2009年10月至2010年9月,取12例PFD患者阴道壁组织块原代培养成纤维细胞,免疫细胞化学法鉴定,绘制生长曲线,Flexercell4000系统加载力,卸载后加入17β-E2,RT-PCR检测Ⅰ型胶原(ColⅠ)、Ⅲ型胶原(ColⅢ)、基质金属蛋白酶-1(MMP-1)和金属蛋白酶组织抑制剂-1(TIMP-1)的表达。结果 (1)应力组ColⅠ、MMP-1表达较静力组明显增强,ColⅢ、TIMP-1表达明显减弱(P<0.01)。(2)17β-E2干预后,ColⅠ、ColⅢ、TIMP-1表达较对照组增强,MMP-1表达减弱(P<0.05)。结论 (1)应力改变了体外培养成纤维细胞的胶原代谢状态。(2)17β-E2部分逆转了应力后成纤维细胞的低反应性。
Objective To investigate the mechanical response of vaginal wall fibroblasts and the effect of 17β-estradiol (E2) on the mechanical response of pelvic floor dysfunction (PFD) in vitro. Methods From October 2009 to September 2010, 12 primary PFD patients with primary vaginal wall tissue fibroblasts were cultured in vitro. Immunocytochemistry was used to identify the growth curves. The growth curve, Flexercell4000 system loading force and unloading The expression of ColⅠ, Col Ⅲ, MMP-1 and TIMP-1 were detected by RT-PCR after addition of 17β-E2. Results (1) The expression of ColⅠand MMP-1 in stress group was significantly higher than that in static group, and the expressions of Col Ⅲ and TIMP-1 were significantly decreased (P <0.01). (2) After 17β-E2 intervention, the expression of ColⅠ, ColⅢ and TIMP-1 increased compared with the control group, while the expression of MMP-1 decreased (P <0.05). Conclusions (1) Stress changes the collagen metabolism status of cultured fibroblasts in vitro. (2) 17β-E2 partially reversed the low reactivity of fibroblasts after stress.
