目的观察人重组酸性成纤维细胞生长因子(aFGF)对原代培养鼠胚神经上皮细胞分化及受体表达的影响。方法应用神经细胞原代培养、免疫细胞化学及图像分析。结果aFGF可使神经元特异性烯醇化酶(NSE)阳性细胞数增加,胞体增大,细胞培养第7天其作用最明显;NSE免疫阳性着色分布于神经细胞胞质和突起中。在培养第3,7,10天,aFGF可促进其受体flg蛋白的表达。结论aFGF可诱导鼠胚神经上皮细胞分化及受体表达
Objective To observe the effect of human recombinant human acidic fibroblast growth factor (aFGF) on the differentiation of neural epithelial cells and the expression of its receptor in primary cultured mouse embryos. Methods Primary culture of neural cells, immunocytochemistry and image analysis were applied. Results aFGF increased the number of neuron-specific enolase (NSE) -positive cells and increased the body cell body. The effect of aFGF on the 7th day was the most obvious. NSE immunoreactive staining distributed in the cytoplasm and protrusions of neurons. On days 3, 7 and 10, aFGF promoted the expression of its receptor flg protein. Conclusion aFGF can induce neural epithelial differentiation and receptor expression in mouse embryos
