从香蕉果实中分离获得1个TIFY亚族的转录因子基因,命名为MaTIFY1。该基因含有一个681 bp的ORF,编码一条长度为227 aa,分子量为24.97 kD,等电点为7.85的氨基酸多肽链。MaTIFY1定位于细胞核,并且具有转录激活活性。荧光定量PCR分析表明,MaTIFY1随着香蕉果实成熟进程表达明显增强,并且外源丙烯处理可诱导其表达。此外,MaTIFY1启动子活性受乙烯利诱导激活,进一步表明MaTIFY1可能参与香蕉果实成熟的调控。原核表达并纯化了MaTIFY1重组蛋白。
One TIFY subfamily transcription factor gene was isolated from banana fruit and named MaTIFY1. The gene contains a 681 bp ORF encoding a polypeptide chain of amino acids with a length of 227 aa, a molecular weight of 24.97 kD and an isoelectric point of 7.85. MaTIFY1 is localized to the nucleus and has transcriptional activation activity. Fluorescence quantitative PCR analysis showed that the expression of MaTIFY1 was enhanced with the progress of banana fruit ripening, and the exogenous propylene treatment induced its expression. In addition, the activation of MaTIFY1 promoter was induced by ethephon, further indicating that MaTIFY1 may be involved in the regulation of banana fruit ripening.
