目的:建立RP-HPLC法同时测定五味首乌片中多指标成分的含量。方法:采用Akasil C18(4.6mm×250mm,5μm)色谱柱,以乙腈(A)-0.4%磷酸(B)为流动相,梯度洗脱(0min,30%A;15min,50%A;20~30min,75%A;35~40min,30%A),流速1.0mL·min-1,检测波长270nm,柱温30℃,进样量20μL;供试品溶液制备采用溶剂分级萃取。结果:淫羊藿苷、大黄素、大黄素甲醚线性范围分别为1.44~7.18,1.28~6.39,0.690~3.45μg;平均加样回收率(n=6)分别为101.6%,103.0%,99.55%。结论:本方法快速、灵敏,重现性好,适合于五味首乌片中淫羊藿苷、大黄素和大黄素甲醚含量测定。
Objective: To establish an RP-HPLC method for the simultaneous determination of multi-indicator components in Wuweishouwu tablets. METHODS: Akasil C18 (4.6 mm×250 mm, 5 μm) column was used with acetonitrile (A)-0.4% phosphoric acid (B) as mobile phase with gradient elution (0 min, 30% A; 15 min, 50% A; 20~ 30min, 75% A; 35 ~ 40min, 30% A), flow rate 1.0mL · min-1, detection wavelength 270nm, column temperature 30 °C, injection volume 20μL; preparation of the test solution using solvent fractionation extraction. RESULTS: The linear ranges of icariin, emodin, and rhubarb were 1.44 to 7.18, 1.28 to 6.39, and 0.690 to 3.45 μg, respectively. The average recovery (n=6) was 101.6%, 103.0%, and 99.55, respectively. %. Conclusion: This method is rapid, sensitive and reproducible. It is suitable for the determination of icariin, emodin and emodin in Wuweishouwu tablets.
