①目的探讨先锋霉素Ⅵ玻璃体腔内注射的安全剂量。②方法以不同浓度的先锋霉素Ⅵ(100,200,250,300,400μg)行兔眼玻璃体腔内注射,用酶组织化学染色法测定注射后不同时间(1,3,7d)视网膜琥珀酸脱氢酶(SDH)和乳酸脱氢酶(LDH)活性的变化,并观察对视网膜组织结构及超微结构的影响。③结果随用药剂量增大,视网膜内SDH与LDH活性均逐渐降低;各剂量组视网膜内SDH与LDH活性分别于术后3d和1d降至最低,然后开始恢复,至手术7d后100和200μg组恢复至正常,其余剂量组仍低于正常。组织学检查显示100和200μg剂量组视网膜出现水肿,其余剂量组均出现细胞变性坏死、杆锥体脱失等改变。④结论先锋霉素Ⅵ玻璃体腔内注射的安全剂量为200μg.
Objective To explore the safe dose of cephalosporin Ⅵ intravitreal injection. ②Methods Different concentrations of cephamycin Ⅵ (100,200,250,300,400μg) were injected into vitreous cavity of rabbits and the retinal succinic acid was determined by enzymatic histochemical staining at different time points (1, 3, 7 days) Dehydrogenase (SDH) and lactate dehydrogenase (LDH) activity changes, and observe the retina tissue structure and ultrastructure. ③Results SDH and LDH activities in the retina decreased gradually with the increase of the dosage; the activity of SDH and LDH in the retina of each dose group decreased to the lowest at 3d and 1d after operation, and then began to recover. At 100 and 200μg after 7d Returned to normal, the remaining dose group is still lower than normal. Histological examination revealed retinal edema in the 100 and 200 μg dose groups, with alterations in cell degeneration and necrosis and rod pyramidal loss in the remaining dose groups. ④ Conclusion The safe dose of cephalosporin Ⅵ vitreous cavity injection is 200μg.
