大岩桐(Sinningia speciosa)为原产巴西的多年生草本花卉。品种多、观赏价值高。利用播种、分球等方法繁殖系数低。我们自1987年开始利用愈伤组织进行快速繁殖。取正在生长的大岩桐幼嫩叶常规消毒,切成直径0.5厘米的小方块,接种在MS附加BA2毫克/升+NAA0.01毫克/升+2,4-D 0.1毫克/升培养基上,50—60天后形成白色致密的愈伤组织。将其转移到附加BA 2毫克/升+NAA 0.01毫克/升的MS培养基上,20天后生出绿芽。在大岩桐继代繁殖中,从无菌苗的下部取1—2对叶移植到培养基上,经一个半月的培养不经愈伤组织,可直接分化形成0.2—1厘米长的不定芽。分化芽的部位主要在叶腹面中脉、侧脉上。
