目的 探讨尿激酶型纤溶酶原激活因子 (uPA)基因在人脑胶质瘤中的表达及临床意义。方法 采用Northern印迹分子杂交和免疫组化方法检测 43例人脑胶质瘤和 5例正常脑组织uPAmRNA的蛋白表达 ,并与临床生物学参数进行综合分析。结果 上述组织或细胞均可表达 2 5Kb的uPAmRNA转录物 ,高级别胶质瘤较低级别和正常脑组织uPAmRNA表达水平明显增高 (P <0 0 1) ,低级别胶质瘤与正常脑组织的表达无显著差异 (P >0 0 5 ) ;术后 18个月内复发和生存期 <3年者uPAmRNA表达显著高于术后 18个月无复发及生存期 >3年者 (P <0 0 1) ,uPAmRNA表达水平与胶质瘤微血管数量呈显著正常相关 (r=0 5 6 ,P <0 0 1) ,与患者性别、年龄及肿瘤大小无显著相关 (P>0 0 5 )。uPA蛋白主要分布在胶质母细胞瘤、间变性星形细胞瘤的瘤细胞及内皮细胞 ,低级别胶质瘤较少。结论 uPA基因表达可能参与胶质瘤的侵袭、血管生成及恶性演变 ,并对胶质瘤的复发和预后有一定意义
Objective To investigate the expression of urokinase-type plasminogen activator (uPA) in human glioma and its clinical significance. Methods The expressions of uPA mRNA in 43 human gliomas and 5 normal brain tissues were detected by Northern blot hybridization and immunohistochemistry. The expressions of uPA mRNA and clinical biological parameters were analyzed. Results The above tissues or cells expressed uPA mRNA transcripts of 25Kb, the expression levels of uPA mRNA in high grade gliomas at lower and normal levels were significantly increased (P <0.01), and those in low grade gliomas and normal brain tissues (P> 0.05). The expression of uPA mRNA in patients with recurrence and survival <3 years after operation was significantly higher than those without recurrence and with survival of> 3 years after 18 months (P <0 05) 1). The expression level of uPA mRNA was significantly and positively correlated with the number of glioma microvessels (r = 056, P <0.01), but not with the gender, age and tumor size (P> 0.05). uPA protein is mainly distributed in glioblastoma, anaplastic astrocytoma of tumor cells and endothelial cells, low-grade glioma less. Conclusion The expression of uPA gene may be involved in the invasion, angiogenesis and malignant transformation of glioma, and has some significance for the recurrence and prognosis of gliomas
