目的:探讨家兔光气中毒后肝脏自由基损伤情况及研究紫外线照射充氧自血回输(UBIO)的保护作用。方法:健康家兔50只随机分为5组,每组10只,雌雄不拘,包括正常对照组、中毒组、常规治疗组、UBIO治疗组及UBIO联合常规治疗(联合治疗)组,连续观察7d,记录家兔存活率,检测肝组织中总抗氧化力(T-AOC)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px),还原性谷胱甘肽(GSH)活性及丙二醛MDA含量。结果:家兔光气中毒后肝脏T-AOC、SOD、GSH-Px、GSH、CAT活性明显低于正常对照组(P<0.01),而MDA含量明显高于正常对照组(P<0.01);在经UBIO治疗或联合治疗后,肝组织中T-AOC、SOD、GSHPx、GSH、CAT活性明显高于中毒组和常规治疗组(P<0.01),而MDA含量明显低于中毒组和常规治疗组(P<0.01)。联合治疗组肝组织中T-AOC、SOD、GSH、GSH-Px、CAT活性与UBIO治疗组差异无统计学意义(P>0.05),而MDA含量明显低于UBIO治疗组(P<0.01)。结论:家兔光气中毒后可引起肝脏自由基损伤,UBIO治疗有明显的保护作用,因而可以用于急性光气中毒的治疗或辅助治疗。
Objective: To investigate the liver injury induced by phosgene in rabbits and the protective effect of UBIO after UVB irradiation. Methods: Fifty healthy rabbits were randomly divided into five groups (n = 10 each), male and female, including normal control group, poisoning group, routine treatment group, UBIO treatment group and UBIO combined with conventional treatment (combination therapy) , The survival rate of rabbits was recorded and the total antioxidant capacity (T-AOC), superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px ), Glutathione (GSH) activity and malondialdehyde MDA content. Results: The activities of T-AOC, SOD, GSH-Px, GSH and CAT in rabbits were significantly lower than those in normal control group (P <0.01), while the contents of MDA in rabbits were significantly higher than those in normal control group (P <0.01). The activity of T-AOC, SOD, GSHPx, GSH and CAT in liver tissue of UBIO group were significantly higher than that of poisoning group and conventional treatment group (P <0.01), while the content of MDA was significantly lower than that of poisoning group and routine treatment Group (P <0.01). The activities of T-AOC, SOD, GSH, GSH-Px and CAT in the hepatic tissue of the combination therapy group were not significantly different from those of the UBIO treatment group (P> 0.05), but the content of MDA was significantly lower than that of the UBIO treatment group (P <0.01). Conclusion: Rabbits phongene poisoning can cause liver free radical damage, UBIO treatment has a significant protective effect, which can be used for the treatment of acute phosgene poisoning or adjuvant therapy.
