目的:研究骨巨细胞瘤(giantcelltumorofbone,GCT)组织尿激酶型纤溶酶原激活剂(urokinase-typeplasminogenactivator,u-PA)系统mRNA的表达与GCT病理分级和复发的关系。方法:用原位杂交技术检测42例GCT组织u-PA,u-PA受体(u-PAR)及其抑制因子2(PAI-2)mRNA的表达。结果:①u-PA,u-PAR,PAI-2mRNA在GCT组织多核巨细胞(MGC)的阳性率分别为64.3%,71.4%和40.5%;单核基质细胞(MC)的阳性率分别为54.8%,45.2%和9.5%。②Ⅱ、Ⅲ级和复发组GCT的MGCu-PAmRNA与MCu-PA,u-PARmRNA的表达率均明显高于Ⅰ级(χ2分别为8.00,7.82,6.81,P<0.05)和无复发组(χ2分别为5.49,5.54,9.84,P<0.05);③GCT组织两种细胞间的u-PAmRNA表达(P<0.05)、MGCu-PAR与PAI-2mRNA的表达间、MCu-PA与u-PARmRNA的表达间均呈正相关(χ2分别为7.20,8.19,P<0.05);MGCPAI-2mRNA的阳性率明显高于MC的阳性率(χ2=10.73,P<0.05)。结论:GCT组织u-PA,u-PARmRNA的表达,尤其是在MC中的表达,与骨巨细胞瘤恶性程度密切相关。
Objective: To investigate the relationship between urokinase-typeplasminogen activator (u-PA) mRNA expression and GCT pathological grading and recurrence in giant cell tumor of bone (GCT). Methods: The expressions of u-PA, u-PA receptor (PAI-2) and u-PA mRNA in 42 GCT tissues were detected by in situ hybridization. Results: ① The positive rates of u-PA, u-PAR and PAI-2 mRNA in GCT tissues were 64.3%, 71.4% and 40.5%, respectively. The positive rates of mononuclear stromal cells were 54.8% , 45.2% and 9.5% respectively. ② The expression rates of MGCu-PA and MCu-PA and u-PARmRNA in grade Ⅱ, grade Ⅲ and relapsed GCT were significantly higher than those in grade Ⅰ (χ2 = 8.00,7.82,6.81, P <0.05, respectively) (5.49,5.54,9.84, P <0.05). The expression of u-PA mRNA between MGCu-PAR and PAI-2mRNA, (Χ2 = 7.20,8.19, P <0.05). The positive rate of MGCPAI-2 mRNA was significantly higher than that of MC (χ2 = 10.73, P <0.05). Conclusion: The expression of u-PA and u-PARmRNA in GCT tissues, especially in MC, is closely related to the malignant degree of giant cell tumor of GCT.
