目的探讨肿瘤坏死因子-α(TNF-α)基因单核苷酸多态性(SNP)与胃腺癌及幽门螺杆菌(Hp)感染胃腺癌发生发展的相关性。方法采用基因芯片技术检测130例胃腺癌患者和142名健康对照人群中 TNF-α-238G/A、-308G/A 位点多态性。同时应用酶联免疫吸附试验(ELISA)测定了其血清中 Hp-IgG/IgM/IgA 型抗体浓度。结果感染 Hp 胃腺癌患者的阳性率明显高于感染 Hp 的对照组(P<0.01)。TNF-α-238GA 基因型和 A 等位基因频率,在胃腺癌组明显高于对照组(P<0.01,P<0.01),在 Hp 阳性胃腺癌组明显高于 Hp 阴性胃腺癌(P<0.05,P<0.01)。未见 TNF-α其他位点的 SNP 与胃腺癌组或 Hp 阳性胃腺癌组有任何相关性。结论 TNF-238GA 基因型及其等位基因 A 与胃腺癌或感染 Hp 的胃腺癌易感性相关。
Objective To investigate the association between single nucleotide polymorphism (SNP) of tumor necrosis factor-α (TNF-α) gene and the development of gastric adenocarcinoma and Helicobacter pylori (Hp) infection. Methods TNF-α-238G / A and -308G / A polymorphisms in 130 patients with gastric adenocarcinoma and 142 healthy controls were detected by gene chip technique. Serum Hp-IgG / IgM / IgA antibody concentrations were also determined by enzyme linked immunosorbent assay (ELISA). Results The positive rate of Hp gastric adenocarcinoma was significantly higher than that of Hp infected control group (P <0.01). The frequencies of TNF-α-238GA genotype and A allele in gastric adenocarcinoma group were significantly higher than those in control group (P <0.01, P <0.01), and were significantly higher in Hp positive gastric adenocarcinoma group than in Hp negative gastric adenocarcinoma group (P <0.05) , P <0.01). No SNP found in other sites of TNF-α had any correlation with gastric adenocarcinoma group or Hp positive gastric adenocarcinoma group. Conclusion The genotype of TNF-238GA and its allele A are associated with the susceptibility to gastric adenocarcinoma or gastric adenocarcinoma infected with Hp.
