目的 :建立人血浆中盐酸多奈哌齐的HPLC MS法。方法 :血样经饱和NaHCO3 碱化后用乙酸乙酯提取 ,进行HPLC MS分析 ,色谱柱为HypersilODS(5μm ,2 50mm× 4 6mm) ,流动相为甲醇 水 三乙胺 冰醋酸 (70∶3 0∶0 3∶0 3 ) ,内标为盐酸非洛普 ,检测离子为m /z 3 80 (多奈哌齐 )、m/z 3 4 4 (内标 ) ,裂解电压为 12 0V。结果 :在 0 2~ 2 0ng/ml范围内多奈哌齐与内标峰面积比值与浓度线性关系良好 (r =0 9996) ,最低可定量浓度为 0 1ng/ml,提取回收率为 87 52 %~ 90 91%。结论 :本实验建立的分析方法灵敏、准确、简便
Objective: To establish HPLC MS method of donepezil hydrochloride in human plasma. Methods: The blood samples were alkalified by saturated NaHCO3, extracted with ethyl acetate and analyzed by high performance liquid chromatography (HPLC). The column was Hypersil ODS (5μm, 250mm × 4.6mm) and the mobile phase was methanol water triethylamine glacial acetic acid (70: 3 0: 0 3:0 3), the internal standard was felop hydrochloride, the detection ion was m / z 3 80 (donepezil), m / z 34 4 (internal standard) and the lysis voltage was 12 0V. Results: The linear relationship between the ratio of donepezil and internal standard peak area was good (r = 0 9996) in the range of 0 2 ~ 20 ng / ml, the lowest quantifiable concentration was 0 1 ng / ml and the extraction recovery was 87 52% ~ 90 91%.
