目的 探讨ICAM -1在胃癌细胞、胃癌患者腹腔液和血浆中的表达及水平与胃癌TNM分期之间的关系。方法 3 5例胃癌患者采用流式细胞仪检测肿瘤细胞表面ICAM -1水平 ;用ELISA方法定量检测腹水及血浆中可溶性ICAM -1水平及其细胞阳性表达率。结果 肿瘤细胞表面ICAM -1的表达较对照组明显升高 ,Ⅲ期显著高于Ⅱ期 (P <0 0 5 ) ,淋巴结转移组和无转移组间有显著差异 (P <0 0 5 )。两组Ⅰ期与Ⅱ期间sICAM -1水平无明显差异 (P >0 0 5 ) ,Ⅲ期明显高于Ⅱ期 (P <0 0 5 ) ,有淋巴结转移者明显高于无转移者 (P <0 0 5 )。手术切除肿瘤后 ,血浆中sICAM -1的水平下降 ,并与TMN分期有关。结论 肿瘤细胞表面ICAM -1阳性率及腹水和血浆中可溶性ICAM -1浓度随着胃癌TNM分期增高而有上升趋势 ,且与淋巴结转移有关 ,可作为判断肿瘤发展程度、转移状况以及预后的指标之一。
Objective To investigate the expression of ICAM-1 in peritoneal fluid and plasma of patients with gastric cancer and gastric cancer and its relationship with the TNM staging of gastric cancer. Methods 35 ICAM-1 levels on the surface of tumor cells were detected by flow cytometry in 35 gastric cancer patients. The levels of soluble ICAM-1 and the positive rate of ICAM-1 in ascites and plasma were detected by ELISA. Results The expression of ICAM - 1 on the surface of tumor cells was significantly higher than that of the control group. The expression of ICAM - 1 was significantly higher in stage Ⅲ than that in stage Ⅱ (P <0.05). There was a significant difference (P <0.05) between ICAM - 1 expression in lymph node metastasis and without metastasis. There was no significant difference in the level of sICAM-1 between stage I and stage II (P> 0.05), stage III was significantly higher than stage II (P <0 05), with lymph node metastasis was significantly higher than those without metastasis (P < 0 0 5). After surgical resection of the tumor, plasma levels of sICAM-1 are reduced and are related to TMN staging. Conclusions The positive rate of ICAM-1 on the surface of tumor cells and the concentration of soluble ICAM-1 in ascites and plasma are increasing with the increase of TNM staging of gastric cancer, which may be related to lymph node metastasis and may be used as an indicator of tumor progression, metastasis and prognosis one.
