目的:探讨化瘀消瘤方含药血清对肺癌细胞周期的作用。方法:制备高、中、低剂量含药血清、西药血清和对照血清,作用于人肺癌细胞。MTT法观察血清对细胞增殖的作用,流式细胞仪进行细胞周期时相分析。结果:高剂量含药血清组与对照血清组之间的吸光度差异有统计学意义(P<0·01),中剂量血清组、高剂量血清组的S期细胞所占比例与对照血清组之间的差异有统计学意义(中剂量血清组P<0·05,高剂量血清组P<0·01)。结论:本实验提示化瘀消瘤方含药血清可能是通过干扰细胞DNA合成而抑制肺癌细胞的增殖,为该药的临床应用提供了科学的实验依据。
Objective: To investigate the effect of serum containing Huatan Xiaoliu Recipe on cell cycle of lung cancer. Methods: High-, medium- and low-dose drug-containing serum, western medicine serum and control serum were prepared and applied to human lung cancer cells. The effect of serum on cell proliferation was observed by MTT method, and the cell cycle was analyzed by flow cytometry. Results: There was a statistically significant difference in the absorbance between the high-dose drug-containing serum group and the control serum group (P<0.01), the proportion of S-phase cells in the middle-dose serum group and the high-dose serum group and the control serum group. The difference between the two groups was statistically significant (medium dose P<0.05, high dose P<0.01). Conclusion: This experiment suggests that the serum containing Huatan Xiaoliu Recipe may inhibit the proliferation of lung cancer cells by interfering with cell DNA synthesis, and provides scientific experimental basis for the clinical application of this drug.
