酵母双杂合系统是在酵母细胞中研究蛋白- 蛋白相互作用的新技术。应用该技术,以丙型肝炎病毒(HCV) 非结构蛋白5A(NS5A) 为“诱饵”,筛检了人肝癌细胞HepG2 来源的cDNA文库,获得了7 个NS5A 结合蛋白的基因克隆。报道了其中两个阳性克隆( # 2 、# 16) 的结果,并对“人核小体组装蛋白相关蛋白”(Human nucleosomeassembly protein -related protein ,hNRP) 和“载脂蛋白A-I”(Apolipoprotein A- I) 与NS5A结合的生物学意义进行了讨论。
The yeast two-hybrid system is a new technique for studying protein-protein interactions in yeast cells. Using this technique, a HepG2-derived cDNA library of human hepatocellular carcinoma cells was screened with the hepatitis C virus (NS5A) non-structural protein 5A (“NS5A”) as bait, and seven NS5A binding protein gene clones were obtained.
