目的观察肿瘤坏死因子(TNF-α)对豚鼠耳蜗血管纹微血管内皮细胞纤维肌动蛋白(F-actin)的影响,为研究豚鼠耳蜗血管纹微血管内皮细胞通透性改变的机制提供依据。方法体外分离培养豚鼠耳蜗血管纹微血管内皮细胞,用0.05、0.1、0.2ng/ml浓度的TNF-α作用90min后,用免疫荧光激光共聚焦扫描显微镜法测定其中各组的F-actin含量变化,对照组为空白对照。结果TNF-α能使耳蜗血管纹微血管内皮细胞的F-actin含量明显减少(P<0.01),且TNF-α的浓度越高F-actin的含量降低越明显。结论通过TNF-α的作用能够改变豚鼠耳蜗微血管内皮细胞内F-actin密度及含量。
Objective To investigate the effect of tumor necrosis factor-α (TNF-α) on the fibroin actin (F-actin) of guinea pig cochlear stria vascularis microvascular endothelial cells and to provide a basis for studying the mechanism of permeability changes of guinea pig cochlear stria microvascular endothelial cells. METHODS: Cultured guinea pig cochlea stria vascularis microvascular endothelial cells in vitro were treated with TNF-α at concentrations of 0.05, 0.1, 0.2 ng / ml for 90 min. The changes of F-actin in each group were detected by immunofluorescence confocal laser scanning microscopy. Control group as a blank control. Results TNF-α significantly decreased the content of F-actin in the microvascular endothelial cells of the cochlear stria vascularis (P <0.01). The higher the concentration of TNF-α, the more significant the decrease of F-actin was. Conclusion The density and content of F-actin in cochlear microvascular endothelial cells of guinea pigs can be changed by the action of TNF-α.
