目的探讨子宫胎盘缺血(RUPP)改良手术方法制作妊娠高血压大鼠(Spragu-Dawley)模型的可行性。方法将30只SD妊娠大鼠随机分成3组:假手术组、RUPP经典手术组(于孕14d手术,在腹主动脉、双侧卵巢动脉置入银夹)、RUPP改良手术组(于孕14d手术,在腹主动脉、单侧卵巢动脉置入银夹),每组10只。结果孕19d时,与假手术组比较,RUPP经典手术、RUPP改良手术组收缩压[(157.4±11.8)、(161.4±6.3)比(117.0±9.4)mm Hg]、舒张压[(122.6±8.1)、(123.3±7.7)比(100.7±4.4)mm Hg]、心率[(398.4±10.1)、(400.5±13.1)比(383.9±11.5)次/min]、尿蛋白[(18.1±3.7)、(17.8±3.2)比(8.4±1.9)μg/μL]明显增高(均P<0.05);母鼠体质量增加值(孕19d体质量-孕14d体质量)[(29.6±4.9)、(42.7±6.9)比(79.4±8.0)g]、仔鼠存活数[(1.2±0.8)、(3.1±0.8)比(12.0±1.6)只]、仔鼠体质量[(1.8±0.1)、(1.7±0.2)比(2.6±0.1)g]、母鼠存活率(50%、80%比100%)、胎盘总超氧化物歧化酶活力[(98.2±7.0)、(99.2±3.6)比(145.0±3.5)U/mg pro]下降(均P<0.05)。与RUPP经典手术组比较,RUPP改良手术组母鼠体质量增加值、仔鼠存活数、母鼠存活率增高(均P<0.05),而血压、尿蛋白、仔鼠体质量差异无统计学意义(均P>0.05)。结论 RUPP诱发妊娠高血压大鼠模型是研究妊娠高血压的良好模型。RUPP改良手术诱发妊娠高血压效果与RUPP经典手术相似。
Objective To investigate the feasibility of using the method of uterine placental ischemia (RUPP) to improve the pregnancy model of hypertensive rats (Spragu-Dawley). Methods Thirty SD pregnant rats were randomly divided into three groups: sham operation group, RUPP classic operation group (14 days pregnancy operation, silver clip in abdominal aorta and bilateral ovary artery), RUPP modified operation group Surgery, in the abdominal aorta, unilateral ovarian artery placed silver clip), each group of 10. The body weight gain (body weight at 19 days pregnant and pregnancy 14 days) [(29.6 ± 4.9) and (6.9 ± 0.7) g / (6.9 ± 0.9) vs (79.4 ± 8.0) g], [(1.2 ± 0.8) vs (3.1 ± 0.8) vs (12.0 ± 1.6) (0.2 ± 0.2) g / (2.6 ± 0.1) g], the survival rate of the female rats (50%, 80% vs 100%), the total superoxide dismutase activity of the placenta [(98.2 ± 7.0), (99.2 ± 3.6) ± 3.5) U / mg pro] (all P <0.05). Compared with the RUPP classic operation group, the body weight gain, surviving pups survival rate and survival rate of dams in RUPP improved group (all P <0.05), but there was no significant difference in blood pressure, urine protein and pups weight (All P> 0.05). Conclusion RUPP induced hypertensive rat model of pregnancy is a good model of pregnancy-induced hypertension. RUPP modified surgery to induce pregnancy-induced hypertension is similar to RUPP classic surgery.
