目的探讨硒对氟诱导大鼠肾小管上皮(NRK-52E)细胞凋亡及caspase-3和caspase-8 mRNA表达的影响。方法取处于对数生长期的肾小管上皮细胞,分别加入0(对照)、5、20 mg/L Na F溶液和(或)17.1、34.2μg/L亚硒酸钠溶液继续培养0、24、48、72、96 h,采用MTT法检测细胞存活率,采用流式细胞仪和Real-time PCR法分别检测染毒48 h时的细胞凋亡率和凋亡相关因子caspase-3和caspase-8 mRNA的表达水平。结果与对照组相比,20 mg/L Na F染毒肾小管上皮细胞存活率均较低,凋亡率较高;与相同剂量Na F染毒组比较,硒干预组细胞存活率均较高,各浓度Na F+17.1μg/L亚硒酸钠染毒组的细胞凋亡率较低。与对照组相比,Na F染毒组肾小管上皮细胞中caspase-3和caspase-8 mRNA的表达水平均较高;与相同剂量Na F染毒组比较,20 mg/L Na F+各浓度亚硒酸钠组细胞中caspase-3和caspase-8 mRNA的表达水平较低。结论一定浓度的硒可能通过下调凋亡相关因子caspase-3和caspase-8的表达来拮抗细胞凋亡。
