目的研究胃癌(gastriccancer,GC)中环氧合酶-2(cyclooxygenase-2,COX-2)、核因子-κB(nuclearfactorκappaB,NF-κB)的表达规律与病理特征的关系。方法采用免疫组织化学染色法(SABC法)和末端脱氧核苷酸转移酶(TDT)介导的dUTP缺口标记法(TUNEL)检测15例正常胃黏膜(normalgastricmucosa,NGM)和40例GC组织中COX-2、NF-κBp65表达情况。结果GC中COX-2、NF-κBp65的阳性表达分别为67.5%、62.5%,NGM组则为13.3%、6.70%,GC组和NGM组差别有显著性(P<0.01)。COX-2、NF-κBp65在低分化GC中表达显著高于高分化型,且后者还与GC病期有关(P<0.05)。COX-2、NF-κBp65与GC淋巴结转移、血管浸润均密切相关(P<0.05)。结论COX-2、NF-κB的表达水平对评价GC的恶性程度、有无转移和预后有一定参考价值。
Objective To investigate the relationship between cyclooxygenase-2 (COX-2) and nuclear factor-κB (NF-κB) expression and pathological features in gastric cancer. Methods Immunohistochemical staining (SABC) and terminal deoxynucleotidyl transferase (TDT) -mediated dUTP nick labeling (TUNEL) were used to detect the expression of COX in 15 normal gastric mucosa (NGM) and 40 GC tissues -2, NF-κBp65 expression. Results The positive expressions of COX-2 and NF-κBp65 in GC were 67.5% and 62.5% respectively, while those in NGM group were 13.3% and 6.70% respectively. There was a significant difference between GC group and NGM group (P <0.01). The expression of COX-2 and NF-κBp65 in poorly differentiated GC was significantly higher than that in well differentiated ones, and the latter was also related to the stage of GC (P <0.05). COX-2 and NF-κBp65 were closely related to lymph node metastasis and vascular invasion (P <0.05). Conclusions The expression of COX-2 and NF-κB may be valuable in evaluating the malignancy, metastasis and prognosis of GC.
