目的:研究单侧输尿管梗阻(UUO)大鼠肾组织p27的表达,同时观察TGF-β mRNA在各组的变化,探讨UUO模型中TGF-β与p27的关系。方法:60只SD大鼠随机分成假手术组(SOR)、UUO模型组。于术后第7、14、21天分别处死各组大鼠10只。用HE染色动态观察肾脏病理变化,免疫组织化学法测定p27的蛋白表达及动态变化,RT-PCR法测定p27 mRNA和TGF-β mRNA的水平。结果:SOR组肾小管上皮细胞p27蛋白及肾皮质p27 mRNA的表达均强,UUO组随着间质纤维化程度的加重,p27的表达逐渐减弱,而TGF-β mRNA随着肾间质纤维化程度的加重,其表达呈上升趋势;UUO组TGF-β mRNA与p27 mRNA成负相关。结论:p27参与了UUO大鼠肾间质纤维化的发病过程;UUO大鼠肾间质纤维化模型中TGF-β促纤维化机制可能与p27的下调相关。
Objective: To investigate the expression of p27 in renal tissue of rats with unilateral ureteral obstruction (UUO), and to observe the changes of TGF-β mRNA in each group and to explore the relationship between TGF-β and p27 in UUO model. Methods: Sixty SD rats were randomly divided into sham operation group (SOR) and UUO model group. On the 7th, 14th and 21st day after operation, 10 rats in each group were sacrificed. The pathological changes of kidney were observed dynamically by HE staining. The protein expression and dynamic changes of p27 were detected by immunohistochemical method. The levels of p27 mRNA and TGF-β mRNA were determined by RT-PCR. Results: The expression of p27 protein in renal tubular epithelial cells and renal cortex p27 mRNA in SOR group was strong. The expression of p27 in UUO group decreased gradually with the degree of interstitial fibrosis, while the expression of TGF-β mRNA in renal tubulointerstitial fibrosis The degree of increase, the expression showed an upward trend; UUO group TGF-β mRNA and p27 mRNA was negatively correlated. Conclusion: p27 is involved in the pathogenesis of renal interstitial fibrosis in UUO rats. The mechanism of TGF-β promoting fibrosis may be related to the down-regulation of p27 in UUO rat model of renal interstitial fibrosis.
