天泰1号对自发阿尔茨海默病鼠相关脑区超氧化物歧化酶和脂褐素的定量研究(英文)

背景:阿尔茨海默病发病的自由基学说以及氧化损害与阿尔茨海默病发病的关系使抗氧化药物研究成为热点,天然中药中的抗氧自由基损伤作用是否可产生对阿尔茨海默病的干预效应?目的:观察天然中药科研制剂天泰1号对自发阿尔茨海默病模型学习记忆功能及氧自由基代谢的影响。设计:以实验动物为研究对象,完全随机分组设计的实验研究。单位:一所市级中西医结合临床研究所。材料:实验在深圳市中西医结合临床研究所二级动物实验室完成。实验动物为昆明种小鼠,清洁级,雄性,13月龄,体质量40~50g,购自中山医科大学实验动物中心(合格证号:粤检证字第95A07号)。干预:实验动物饲养至21月龄,按国际通用标准和方法从老龄动物中筛选阿尔茨海默病小鼠;实验动物随机分为空白对照组、西药对照组、天泰1号小剂量组、天泰1号大剂量组、另设老年学习记忆正常组(简称老年正常组下同)。西药对照组给以甲磺酸双氢麦角毒碱片(Hydergine)0.6mg/kg,天泰1号小、大剂量组分别予天泰1号6.80g/kg及20.41g/kg,连续60d空白对照组和老年正常组均灌以等量双蒸水。主要观察指标:用跳台实验检测学习记忆成绩;脑组织冰冻切片,免疫组织化学法示大脑皮质及海马超氧化物歧化酶活性;海马CA1区脑组织超薄切片,透射电镜法观察神经元细胞脂褐素;全自动显? Whether the antioxidant free radical damage in natural Chinese medicine can produce Alzheimer’s disease Intervention Effect of Disease Objective: To observe the effect of Tiantai No.1, a natural Chinese medicine scientific research preparation, on learning and memory function and oxygen free radical metabolism in a spontaneous Alzheimer’s disease model. Design: An experimental study with completely randomized groupings designed for experimental animals. Unit: A city-level Institute of Integrated Traditional Chinese and Western Medicine. MATERIALS: Experiments were performed at the Class II Animal Laboratory of the Institute of Integrated Chinese and Western Medicine in Shenzhen. The experimental animals were Kunming mice, clean grade, male, 13 months old, and body weight 40-50 g, purchased from the Experimental Animal Center of Sun Yat-sen University of Medical Sciences (Qualification No.: Guangdong Inspection and Certification No. 95A07). Intervention: The experimental animals were raised to 21 months of age, and Alzheimer’s disease mice were selected from old animals according to internationally accepted standards and methods; experimental animals were randomly divided into blank control group, western medicine control group, and Tiantai No. 1 small dose group. Tiantai No. 1 large-dose group, a separate group of elderly learning and memory normal (referred to as the old normal group below the same). The western medicine control group was given 0.6 mg/kg of Hydrergine mesylate tablets (Hydergine), and the small and high-dose groups of Tiantai No. 1 were given 6.80 g/kg and 20.41 g/kg respectively on Tiantai No. 1 and 60d for consecutive days. The control group and the elderly normal group were equally double distilled water. MAIN OUTCOME MEASURES: The performance of learning and memory was tested by step-down experiments; frozen sections of brain tissue were taken; the activities of superoxide dismutase in cerebral cortex and hippocampus were revealed by immunohistochemical method; ultrathin sections of brain tissue in hippocampal CA1 region; and neuronal cell lipids were observed by transmission electron microscopy.
