目的:建立葛根芩连合剂中葛根素的HPLC含量测定方法。方法:色谱柱:ZORBAX Eclipse XDB C18(4.6 mm×150 mm,5μm),流动相:甲醇-0.2%醋酸水溶液(25∶75),流速:1.0 mL·min-1,检测波长:250 nm,进样量:5μL,柱温:25℃。结果:葛根素的回归方程为y=4214.2x+83.295(r=0.9999),在42.08~420.8 ng范围内线性关系良好;平均加样回收率为97.36%。(n=6),RSD为2.7%。结论:本方法简便可靠,重现性好,可用于葛根芩连合剂的质量控制。
Objective: To establish a method for determination of puerarin in Puerarin. METHODS: The mobile phase consisted of methanol-0.2% aqueous acetic acid (25:75), the flow rate was 1.0 mL · min-1 and the detection wavelength was 250 nm. The mobile phase was ZORBAX Eclipse XDB C18 (4.6 mm × 150 mm, Sample volume: 5 μL, column temperature: 25 ° C. Results: The regression equation of puerarin was y = 4214.2x + 83.295 (r = 0.9999). The linear relationship was good in the range of 42.08-420.8 ng. The average recoveries were 97.36%. (n = 6) with a RSD of 2.7%. Conclusion: The method is simple, reliable and reproducible. It can be used for quality control of Gegenqin Lian Mixture.
