用单细胞培养法,从人胃癌细胞系BGC-823中分离出10个克隆。通过裸鼠致瘤和转移检测,进一步筛选出具有较高转移性的C_1、C_6和低转移的C_5、C_8等4个细胞株。它们的生长能力虽无明显差异,但超微结构显示C_5和C_8细胞表面微绒毛少,而且长度不一,微丝长而粗,排列规则,组装程度较好;C_1和C_6细胞微绒毛多而密集,长度均一,微丝短而粗,呈点状或玫瑰状小体,组装程度差。此外,本研究显示肿瘤细胞内的微丝骨架组装状态,与转移能力似呈负相关。
Ten single clones were isolated from human gastric cancer cell line BGC-823 using single cell culture. Through the tumorigenic and metastatic assays in nude mice, four cell lines, C_1, C_6 with high metastatic potential and C_5 and C_8 with low metastasis, were further screened. Although there was no significant difference in their growth ability, the ultrastructure showed that there were few microvilli on the surface of C_5 and C_8 cells, and the lengths of the microvilli were different. The long and thick microfilaments arranged in a regular way and had a good degree of assembly. C_6 and C_6 cells had many microvilli Dense, uniform length, microfilament short and thick, was a dot or rose-like body, the degree of assembly is poor. In addition, the present study shows that the assembly of actin filaments in tumor cells is negatively correlated with the metastatic potential.
