目的:建立雷丸主要成分的定性与定量分析方法。方法:采用薄层色谱法对雷丸中的麦角甾醇、麦角甾酮进行鉴别。运用可见-紫外分光光度法对雷丸所含的驱绦虫成分雷丸凝集素进行含量测定,以水为提取溶剂,采用Fonlin酚法,测定波长650nm。结果:应用所建立的方法对13批雷丸药材进行定性与定量分析,结果TLC斑点清晰,专属性强,均可检出雷丸的特征斑点;13批雷丸药材雷丸凝集素的含量以牛血清白蛋白计为0.61%~4.23%;方法学研究结果表明,牛血清白蛋白的量在52.76~263.8μg范围内呈良好的线性关系(r=0.9997),平均回收率为98.0%(RSD=1.0%,n=6)。结论:本方法操作简便,重复性好,结果准确可靠,可用于雷丸的定性定量分析,以控制雷丸药材及相关产品的质量。
Objective: To establish a qualitative and quantitative analysis method for the main components of Leiwan. Methods: The ergosterol and ergosterone in Leiwan were identified by TLC. Using visible - UV spectrophotometry Radix pill containing tapeworm ingredient Leiyan lectin content determination, water as the extraction solvent, using Fonlin phenol method, determination of wavelength 650nm. Results: The method was used to establish a qualitative and quantitative analysis of 13 batches of Radix Pill. The results showed that TLC spots were clear and specific, and the characteristic spots of Ray pill could be detected. The content of 13 batches of Radix pill Bovine serum albumin was 0.61% -4.23%. The results of methodological study showed that the amount of bovine serum albumin showed a good linearity (r = 0.9997) in the range of 52.76 ~ 263.8μg with an average recovery of 98.0% (RSD = 1.0%, n = 6). Conclusion: The method is simple, reproducible, and accurate and reliable. It can be used to qualitative and quantitative analysis of Rayleigh pellets to control the quality of Leiwan pellets and related products.
