目的:本实验从mRNA水平研究肾癌组织、癌旁肾组织及正常肾组织中受体酪氨酸激酶EphB4和基质金属蛋白酶-9(MMP-9)的表达规律及二者表达之间的相关性,探讨其在肾癌发生发展过程中的可能作用,为肾癌的诊断、治疗及预后判断提供新的方法和思路。方法:运用半定量RT-PCR技术检测肾癌组织、癌旁肾组织及正常肾组织中EphB4和MMP-9在mRNA水平的表达情况,分析其与肾癌临床病理因素的相关性,探讨其与肾癌细胞增殖、浸润和转移的关系。结果:EphB4和MMP-9在肾癌组织、癌旁肾组织中都表达,且癌旁表达低于肿瘤组织表达,差异有统计学意义(P<0.05)。二者在正常肾组织中几乎不表达(P<0.01)。两者在肾癌组织中的表达丰度与患者的年龄、性别无关(P>0.05),与肿瘤的临床分期、病理分级以及是否淋巴结转移密切相关(P<0.05)。二者在肾癌组织中表达呈明显正相关关系(r=0.631,P<0.01)。结论:EphB4和MMP-9在肾癌的发生、发展及侵袭转移中起非常重要作用,联合检测两者的表达对预测肾癌患者预后、指导治疗有一定临床意义。
AIM: To investigate the expression of EphB4 and MMP-9 in renal cell carcinoma, paracancer, and normal kidney tissue at the mRNA level and the correlation between the expression of EphB4 and MMP-9 To explore its possible role in the development of renal cell carcinoma and to provide a new method and idea for the diagnosis, treatment and prognosis of renal cell carcinoma. Methods: The mRNA expression of EphB4 and MMP-9 in renal cell carcinoma, paracancer renal tissue and normal renal tissue were detected by semi-quantitative RT-PCR. The correlation between EphB4 and MMP-9 mRNA and clinicopathological parameters was analyzed. Renal cell proliferation, invasion and metastasis. Results: The expressions of EphB4 and MMP-9 in renal cell carcinoma and para-cancerous kidney tissues were significantly lower than those in adjacent normal tissues (P <0.05). The two were almost not expressed in normal kidney tissue (P <0.01). The expression abundance in renal cell carcinoma was not related to age and gender (P> 0.05), but closely correlated with clinical stage, pathological grade and lymph node metastasis (P <0.05). There was a positive correlation between the two expression levels in renal cell carcinoma (r = 0.631, P <0.01). Conclusion: EphB4 and MMP-9 play a very important role in the carcinogenesis, development and invasion and metastasis of renal cell carcinoma. The combined detection of EphB4 and MMP-9 has some clinical significance in predicting the prognosis and guiding treatment of patients with renal cell carcinoma.
