目的探讨艾芬地尔(ifenprodil)对大鼠脑缺血再灌注后凋亡诱导因子(apoptosis inducing fac-tor,AIF)表达及神经元凋亡的影响。方法线栓法建立Wistar大鼠大脑中动脉闭塞(MACO)模型,采用原位末端标记法(TUNEL)及免疫组化法分别于6、24、48、72和168h观察假手术组、对照组及艾芬地尔(10mg/kg,ip)组缺血半暗带区神经元凋亡数及AIF阳性细胞数的变化。结果艾芬地尔组大鼠脑组织凋亡细胞数及AIF阳性细胞数均低于对照组(P<0.05)。结论艾芬地尔具有抗凋亡作用,其机理可能系通过阻滞变化[N-甲基-D-天冬氨酸盐(N-Methyl-D-aspartate,NMDA)]受体NR2B亚基,降低兴奋性氨基酸毒性,减少AIF的释放,来抑制AIF介导的细胞凋亡。
Objective To investigate the effect of ifenprodil on the expression of apoptosis inducing fac-tor (AIF) and neuronal apoptosis after cerebral ischemia-reperfusion in rats. Methods The middle cerebral artery occlusion (MACO) model of Wistar rats was established by thread occlusion method. TUNEL and immunohistochemical method were used to observe the changes of MACO in sham operation group, control group and control group at 6, 24, 48, 72 and 168 h The changes of neuronal apoptosis and AIF positive cells in the ischemic penumbra in the group of alprenilone (10mg / kg, ip). Results The number of apoptotic cells and the number of AIF positive cells in the ischemic group were lower than those in the control group (P <0.05). Conclusions Affinisidil has an anti-apoptotic effect and its mechanism may be through the change of block [N-Methyl-D-aspartate (NMDA)] receptor NR2B subunit, Reduce excitatory amino acid toxicity, decrease AIF release, and inhibit AIF-mediated apoptosis.
