目的:筛选具有拮抗CpGODN作用的抗炎中药。方法:将CpGODN包被于生物传感器的生物素化样品池上,测定78种中药水煎液与CpGODN的亲和力;选择具有较高亲和力的中药水煎液,与CpGODN(33μg·ml-1)37℃孵育30min后,再次测定与CpGODN的亲和力。结果:78种中药中,地榆、大黄、藏青果等14种中药含有结合CpGODN的活性成分;鸦胆子、赤芍、地榆等9种中药活性成分含量较高。结论:筛选得到9种活性含量较高的中药,为下一步分离提纯有效成分单体提供了实验依据。
OBJECTIVE: To screen anti-inflammatory Chinese medicines with antagonistic effect on CpGODN. METHODS: CpG ODN was coated on the biosensor biotinylated sample cell and the affinity of 78 Chinese herbal decoctions and CpGODN was determined. The Chinese medicine decoction with higher affinity was selected and CpGODN (33μg·ml-1) 37°C. After incubation for 30 min, affinity to CpGODN was again determined. Results: Among 78 kinds of traditional Chinese medicines, 14 kinds of traditional Chinese medicines such as Radix przewalskii, rhubarb, and Tibetan green fruit contained active ingredients that combined with CpGODN. The contents of 9 active ingredients of Chinese herbs such as Brucea javanica, Radix paeoniae Rubra and Radix paeoniae Rubra etc. were higher. Conclusion: Nine kinds of traditional Chinese medicines with higher activity content were screened, which provided experimental evidence for the separation and purification of active ingredients in the next step.
