目的:研究白鹤冲剂对人脐静脉内皮细胞(HUVEC)中核因子-κB(NF-κB)活化、细胞间黏附分子-1(ICAM-1)表达的影响,以探讨白鹤冲剂治疗血管炎的机制。方法:以细胞 ELISA 法、间接免疫荧光法分别观察白鹤冲剂对 HUVEC 中 NF-κB活化以及 ICAM-1表达的影响。结果:白鹤冲剂能抑制肿瘤坏死因子-α(TNF-α)刺激的 HUVEC 的 NF-κB的活化及 ICAM-1表达。结论:白鹤冲剂可能通过抑制 NF-κB活化从而有效地调控与炎症过程密切相关的黏附分子的基因表达,发挥治疗血管炎的重要作用。
Objective: To study the effects of Baihe Instant Granules on the activation of nuclear factor-κB (NF-κB) and the expression of intercellular adhesion molecule-1 (ICAM-1) in human umbilical vein endothelial cells (HUVEC), and to explore the mechanism of Baihe Granules in treating vasculitis. Methods: The effects of Baihe Granules on NF-κB activation and ICAM-1 expression in HUVEC were observed by cell ELISA and indirect immunofluorescence. Results: Baihe Granules can inhibit the activation of NF-κB and the expression of ICAM-1 in HUVEC stimulated by tumor necrosis factor-α (TNF-α). Conclusion: Baihe Granules may play an important role in the treatment of vasculitis by inhibiting the activation of NF-κB and effectively regulating the gene expression of adhesion molecules closely related to the inflammatory process.
