妊娠期糖尿病孕妇外周血中胎儿游离DNA-SRY的检测

目的探讨孕妇外周血中游离胎儿DNA水平在诊断及预防妊娠期糖尿病(GDM)中的应用价值。方法用血浆DNA抽提法提取30例孕中晚期孕妇外周血中胎儿游离DNA(其中病例组10例,对照组20例),通过实时荧光定量PCR(RQ-PCR)法检测两组胎儿DNA-SRY含量水平。结果14例孕男胎均出现SRY阳性信号,妊娠期糖尿病组孕妇外周血中胎儿游离DNA-SRY水平明显高于对照组,P<0.05(=0.019)。结论妊娠期糖尿病孕妇外周血中游离胎儿DNA水平的异常升高有望成为预测及诊断GDM发病与疾病程度的一项有效指标。 Objective To investigate the value of fetal DNA in peripheral blood of pregnant women in the diagnosis and prevention of gestational diabetes mellitus (GDM). Methods Fetal DNA was extracted from peripheral blood of 30 pregnant women in the second and third trimester of pregnancy (including 10 cases in the case group and 20 cases in the control group) by plasma DNA extraction. Real-time quantitative PCR (RQ-PCR) SRY content level. Results The positive signals of SRY were found in 14 cases of pregnant and maternal fetuses. The fetal DNA-SRY level in pregnant women with gestational diabetes mellitus was significantly higher than that of the control group (P <0.05). Conclusion The abnormal fetal DNA level in peripheral blood of pregnant women with gestational diabetes mellitus is expected to be an effective index to predict and diagnose the incidence and severity of GDM.
