目的:探讨性别差异对脓毒症大鼠血小板活化的影响。方法:40只健康SD大鼠(雌雄各半)随机分为4组(n=10):雄性假手术组、雌性假手术组、雌性脓毒症组、雄性脓毒症组。后两组盲肠结扎穿孔(CLP)法制作脓毒症动物模型,假手术组不行盲肠结扎穿孔。术后24h留取大鼠血液标本,行血小板计数检测,采用ELISA方法检测β血小板球蛋白(β-TG)表达情况,采用放射免疫测定法检测大鼠血清雌二醇(E2)含量。结果:脓毒症雌、雄性大鼠血小板计数均下降,但雌性大鼠明显高于雄性(P<0.01)。脓毒症雌、雄性大鼠血清β-TG表达均升高,但是雌性大鼠表达量明显低于雄性大鼠(P<0.01)。雌、雄性脓毒症大鼠全血血小板计数与相应性别大鼠E2水平显著正相关,血清β-TG水平与相应性别大鼠E2水平显著负相关(P<0.05)。结论:脓毒症的血小板活化存在性别差异,内源性雌激素可能介导了对雌性脓毒症大鼠血小板的保护作用。
Objective: To investigate the effect of gender differences on platelet activation in septic rats. Methods: Forty healthy SD rats (male and female, half) were randomly divided into 4 groups (n = 10): male sham operation group, female sham operation group, female sepsis group and male sepsis group. The latter two groups were made cecal ligation and puncture (CLP) method to make animal models of sepsis. The sham operation group did not cecal ligation and perforation. The blood samples of rats were collected 24h after operation, and the platelet count was measured. The expression of β-TG was detected by ELISA. Serum estradiol (E2) level was measured by radioimmunoassay. Results: The platelet count decreased in female and male sepsis rats, but the female rats were significantly higher than those in male rats (P <0.01). Sepsis male and female rats serum β-TG expression were elevated, but the expression of female rats was significantly lower than male rats (P <0.01). There was a significant positive correlation between whole blood platelet count of female and male sepsis rats and E2 level of corresponding sex rats, and the level of serum β-TG was negatively correlated with the level of E2 of corresponding sex rats (P <0.05). CONCLUSIONS: There are gender differences in platelet activation in sepsis. Endogenous estrogen may mediate the protective effect on platelets in female septic rats.
