目的检测儿童免疫性血小板减少性紫癜(ITP)患者外周血中Th1、Th17、Th22及Treg细胞的比例,以期为ITP的免疫治疗提供实验和理论依据。方法选取来本院就诊的儿童ITP患者48例,同期健康体检儿童30例作为正常对照组。采用流式细胞术检测外周血Th1、Th17、Th22细胞和Treg细胞百分比,分析儿童ITP患者外周血Th1、Th2、Th17、Th22细胞和Treg细胞比例变化与疾病发生的关系。结果 ITP初诊组患者Th2、Th17、Th22细胞百分比高于HC组,而Th1细胞、外周血CD4~+CD25~+Fox P3~+Treg细胞百分比低于正常对照组,差异均有统计学意义(P<0.05)。结论外周血中Th1/Th2、Th17、Th22细胞和Treg细胞比例失衡可能与儿童ITP患者的发病有关。
Objective To detect the proportion of Th1, Th17, Th22 and Treg cells in peripheral blood of children with immune thrombocytopenic purpura (ITP), in order to provide experimental and theoretical evidence for the immunotherapy of ITP. Methods Forty-eight children with ITP were enrolled in this study, and 30 healthy children were selected as normal control group. The percentage of Th1, Th17, Th22 and Treg cells in peripheral blood was detected by flow cytometry. The relationship between Th1, Th2, Th17, Th22 and Treg cells in children with ITP was analyzed. Results The percentage of Th2, Th17 and Th22 cells in newly diagnosed ITP group was higher than that in HC group, while the percentage of CD4 ~ + CD25 ~ + Fox P3 ~ + Treg cells in Th1 and peripheral blood was lower than that in normal control group (P <0.05). Conclusion The imbalance of Th1 / Th2, Th17, Th22 and Treg cells in peripheral blood may be related to the pathogenesis of childhood ITP.
